It's actually from art, not a screencap -- I forget what picture exactly -- but I have it because he does laugh like that! All of once in canon.
Backstory here: Heero was in a coma for a month, after he self-detonated his Gundam under orders. Trowa hauled his unconscious body back to the circus, and has been taking care of him. Heero's just woken up, and Trowa briefs him on the situation and that they're all kind of at loose ends and in hiding and haven't gotten orders for a month, and says something like "I dunno, maybe I should do what you did."
Heero says "Well, if you do, you should know this. It hurts like hell." And Trowa blinks at him, and then cracks up.
(Catherine, overhearing only the laughter, is enormously pleased that Trowa's making friends! I facepalm forever at both the pilots.)
...So, uh, in context I guess that's not the happiest moment ever. But there are only a tiny handful of people who could startle him into laughter like that -- not all of whom have the right sense of humor for it anyway, but they're the ones he lets his guard down enough around -- and also it proves that he is capable of laughing, okay, so I count the icon!
