See here: your pup.
Meanwhile, I try not to eye the Millicanoned best friend from Mal's childhood. Whoops!

I think that Cy misses his old gang, his teammates from Little League and Pop Warner football and his high school teams.

Because... yeah
(Quick survey of 7 pups: three of them have estranged or dead sibling angst)

ETA: And to answer the second part of the question...Flint had an accident involving radiation and so of course gained super powers, like most people exposed to radiation in the MU. He was a bad guy for a long time, most often duking it out with an obnoxious kid who likes to throw webs around. Eventually Flint had an epiphany and decided to turn over a new leaf and be good which lasted for a while but then I think he went bad again.

Tony here used to be best friends with his cousin Angelo, until Angelo got himself brain damage from a car accident and just isn't the same anymore. Weyland misses his closest friend and oldest-younger-brother Egil, his loyal partner in crime, who is very very dead (but that didn't stop him from coming back for a visit). Skaưi's best friend was probably her cousin Ull, and while they still see each other, they've really just grown apart. Butch mainly grew up with his siblings, and while they're fine, he hasn't been back to see them in years on account of the outlaw thing. Al had his younger sister Trudy, who died of neglect in an institution. Fay had friendships back home, but I imagine they were full of heated arguments (she's a very emotional girl) and then she moved to San Francisco and that was that; she does miss her sister Ivy though.
Sqrl depending on his incarnation misses some of his friends from the circus, who are still out there on the road (or in prison). Johnny had some friends back in the old neighborhood, but he left that life behind when he ran away to Joliet, so he doesn't think about them. Joe Bonham's friends likely all got drafted too, though the one he thinks most fondly of is that prostitute back in Shale City who sent him the cufflinks as a graduation present; she's probably still doing her thing.
Loki sorta misses when he and Odin used to be besties, but he would never admit it.

(Yes, rogue geography is a concern. Perhaps not "now", but at some points.)
Or I could make someone up, and make up what happened to them. Maybe pull out... Maria, I think it was? Whoever I said on the "first crush" DE, whenever that was.
Kain... is not all that friendly a person. Cecil is sometimes his friend (and sometimes his archnemesis. It's complicated), but they've never been out of touch with each other.

Really, nothing particularly happened to her; it's just that they live in different (perceptual/metaphorical) worlds. She's, as I mentioned back then, intensely shy, and rather imaginative. Felix is fairly reserved, direct, and concerned with the fate of the world. If he happens to see her when he's in Vale, he makes sure to say hi, but they don't really see things the same way any more.

Ellen wonders sometimes what happened to the people in Vault 101. She tries not to think about Amata, though. That's a serious sore point.

Sunshine's only really close friend from childhood is Aimil, who has been her friend since Rae was seven and Aimil was nine. At the time, Rae was struggling to catch up so she could be admitted into second grade without missing a year (from fleeing her Dad and being hella sick for six months), and Aimil was struggling with the discovery that she was Part-Blood Other. She would say later that she was drawn to Sunshine because she "looked as lost as I felt." They're still friends and keep in touch, often going to local rummage sales and bookstores together. Aimil is a librarian for one of the local libraries (and also for SOF, but shhhhh!), and also named some of Sunshine's signature desserts. :D
Zelgadiss had... hmm. It is likely that Zolf and Rodimus helped train him when he was growing up, and they could be called his childhood friends, though they were much older than he, before Rezo got crazy and red-eyed. They stood by him against Rezo, and died in the battle.

Will counts most of Nottingham this way and I'm not sure of anyone above others.
Sameth doesn't have anyone mentioned in canon. I think growing up his sister was his closest friend since they were the only royalty. He had other friends in Belisaere and at school like Nick but Ellimere's the one who's always been there. Sadly they've grown apart and don't talk like they used to.
Demeter's probably closest to Hephaestus of her siblings but they don't live in the same place so its complicated. Her whole family is rather mixed up.
Moist, I can't think of him having one. He probably got along well with other kids but I don't think there was just one person for him.
Jane's best friend has always been her sister Cassandra and that hasn't changed as they've grown older. I'm sure she has some childhood friends too but I'm not sure.
William's another one where his brother's his best friend. I'm sure there are people he's gotten along with at school but no one in the same way as his brother.
Tumnus I think had a busy childhood that became quite lonely when the Witch took over. So there might have been someone but sadly they probably were killed by the Witch.
The Pirate King, he might have had someone, but I think he's someone who makes friends only when he wants to.

Counting siblings, Lucas still misses his lost twin Claus.

Casey Jones, and April O'Neil.
Raph and Mike miss them both rather spectacularly.

Kendra's childhood friends aren't mentioned and I haven't given her any.
Black Mamba's childhood is a mystery. I haven't been able to find any information about it in canon. But then again, I could peruse all of her canon again and see if anything comes up.
Lockjaw's best friend is Black Bolt.

apparently very intimatelong-term friend. She would say she doesn't care what happened to them, but that would likely be a lie.Dixie has lost three sets of parents, and dearly misses all of her various families. However, she's definitely still close to the girls from the convent, especially the sisters who raised her.

Among my charries, I can only tell about Teja. His childhood friend was Hilde, the grand-daughter of Hildebrand (the original Hilde who the little Gotland OC is named after); she married a Vandal prince and died with him after their people's last battle. Her fate is the subject of the short novel 'Gelimer' by the same author as Teja's canon. She goes into battle along with her husband; and they leave in one chapter expressly to go have sex. It was written ten years after Teja's canon, and something quite fundamental seems to have happened to the author's view about women and sex during those ten years.-
Erm, I digress. Anyway, I have nothing for the others.-
