 How does your pup use humor? Is it the grease to social interactions? Is it a self defence mechanism? It is a shield or a sword? Is it just something fun to play with? Or something else completely?

I bet Brimstone had a sense of humor, once, long ago. I bet it was dry and subtle, but I bet he had one.

He may have had it removed, and if so, he keeps it in a jar for better times.

Dejah is a quick wit, and loves humor. It is the way she measures the spirit of a person. The ease with which they laugh, or can share in a moment of levity, greatly influences how she will interact with someone. The humorless have their uses, but the people she chooses to surround herself with all have a good sense of humor. (I can see her sharing raucous jokes with Tars Tarkas. I can see her giggling at Kantos's puns, even as she slugs him in the arm. I can see her exchanging looks with Sola behind John's back.)

Alex has always had a well-developed sense of humor. One has to in her line of work. If you don't learn to laugh, you die inside a little each time, with every passing case.

Olga has a very droll, very dry sense of humor, and it may be atrophied after so many years of fighting a silent war. But get her drunk and it's there.

Sherlock has a bizarre sense of humor. He does use it as a form of mimicry, to get close to the subjects of his inquiries. But then, he enjoys tormenting John. I also suspect that he and Mrs. Hudson have been known to sit in her kitchen, sharing a joint and giggling their arses off over the stupidest things.

Vlad enjoys a good bit of humor, and it's one of the few shreds of humanity left to him. But it is not his first response, unless the situation warrants it.

John Silver loves a good bit of humor. He uses it as camouflage, to make people think he's just a harmless cook, nothing more. He uses it to diffuse dangerous situations. He uses it in subtle ways as a weapon to cut people down without them even realizing. From his canon:
Long John Silver: I say she's a lovely ship, sir. Well chosen.
Squire Trelawney: Thank you.
Long John Silver: She's got an arse on her that would sit well on any rollicking sea. I always say a man who could choose a good horse could choose a good ship, just by looking at its arse. And you, sir, look like a man who knows his way around a horse's arse.
Vert tends to be a humorous character, but it's a fickle humor that can turn in a heartbeat, as is with all fae. She tends to reflect the person she's interacting with, and if their humor pleases her, then she will share in it.

Earl is about as good-natured as an angel in his position can be expected. While he's not opposed to humor, his experience is that people use it as armor, or to evade deeper questions weighing on their souls. He loves a good belly laugh, but he will not put up with your shit if he thinks you're shining him on. Also, his secret shame is that he loves puns.

I think if Amascut uses any humor, it is mostly to add extra barbs to an insult or it is some snarky observation. The only canonical instances I remember right now is during Diamond in the Rough, particularly when she forces the protagonist to give her the magical diamond called the Kharid-ib, or the Heart of the Sun.

Lady Keli: Finally! After all these centuries… it’s mine!
Leela: Er, centuries?
Lady Keli: In some ways, I’ve been after this heart since the day I was born.
Ozan: Enough with the evil monologue, lady. You’ve got your damned diamond, now hand over the prince!
Lady Keli: And now that I hold it in my hands, it’s so small…
Lady Keli: so hard…
Lady Keli: so cold…
Lady Keli: …
Lady Keli: That explains so much.

Given that her father is god of the sun, this has some … implications.

No ideas about Fairy Fixit's sense of humor other than that her humor might fly over the average person's head.

Duo likes making people laugh. He loves jokes, and pranks. He also uses humor as something to hide behind when he's angry or unhappy. And he can easily make people underestimate him by acting like a twit. Useful all around!

He is still sad that X-23 doesn't think his knock-knock jokes are funny.

Coyote's sense of humor is just as likely to get you killed as not, so it's probably best not to ask her what she thinks is funny. Though she does like atrocious puns.

Donatello and Ambriel are much more staid, and tend toward wry senses of humor with the occasional dip into trolling.

Tommy is a natural wiseass. He likes it when people find him to be funny, being the class clown and all. Even if his jokes are lame or cheesy, he can't resist a punchline or a sarcastic remark. He definitely uses it to break the ice or defuse awkward situations, and in more serious moments, it's a way to cope with all the things he's seen and been through. Lou is the same way, and that's how he and Tommy and the rest of the guys in the firehouse have bonded throughout the years, always wanting to find a way to make each other laugh.

Jim is a Mel Brooks character. Being funny is literally the point of his existence.

Leela probably has a sense of humor, but for the most part she takes life quite seriously.

Kane primarily sees his wit as a weapon he can use to taunt or belittle his opponents. He does appreciate humor in others, but generally he prefers to be laughing at you as opposed to the other way around.

Caius has a sense of humor, but by necessity many of his jokes have to be private - kept to himself and his fellow members of the Blades. He also does make a few affectionate jokes about "sly Uncle Uri," though not to the Emperor's face, of course.

Garyn has a very dry and grim sense of humor, which he primarily uses as a means of coping with how utterly shit the first 26 years of his life have been.

Marge is perpetually good-natured. What good is life without a good laugh now and then, especially at yourself?

I know I'm very late to this party, but...

Ibani uses humor a lot, especially snark. It started as a defense mechanism on Korriban. Snark being an excellent substitute for outright cruelty and a help in convincing the other Sith that she wasn't easy prey. Admittedly, learning how far she could afford to go before getting punished with Force Lightning was...painful.

In game, the Sith Inquisitor snarks all over the place, makes jokes, and generally trolls her enemies. (There's a bonus quest where you corrupt the images in a data archive. The Inquisitor turns the images from Jedi and Republic Senators to Imperial Officers, Sith, and a pole dancer.)
