Sam: Well, not dying in the war is a start, but right now, fulfilling her duty as Mr. Foyle's driver is the main goal, and finding a boyfriend would be nice. She doesn't have a lot of long-term goals, as she's really not sure there's a long-term to be planning for.
Katya: Do what you're good at, try to be a good person.... don't drink too much horrible vodka. It's a simple list, really.
Clara: Do something with your life that's worth doing. Or perhaps, do that thing for someone worth doing it for. Of course, Clara isn't known for her awesome life choices.
Oswin: .... ... Not being a Dalek. Short list.
Jemma: Do what you love, and stay true to who you are. There'll probably be a few extra details on there once she hits canon.
Balthazar: Kill Morgana, save Veronica. ... Oh, wait, in general? Err.... don't be Morgana?
I'll have to do the rest later, time to get to work. :D
Jess once bought into the independently wealthy and famous motif for success. Now she'll consider herself successful when people see her first and not the skrull queen. Once she gets to the Infinity storyline, just seeing the world is still there will be her biggest win. She thinks if she gets another of those Marvel Comics written about her, she'll have made it again.
Hank thinks keeping the US safe is a measure of success. I think he also buys into the getting the girl dream. Helping find and protect other mutants will become his life's work but he's not there yet.
Andrea counts herself successful every time she sees a happy family in Atlanta.
Now with more pups since I've had caffeine and Dead Kennedys to wake me.
Mulan honestly isn't sure what to say aside from keeping your word and remaining true to your duty. (A very quiet voice is saying, "Be honest with Aurora about her feelings.")
Brimstone''s answer would be a spoiler. Let's just say a chance for life to grow.
Quin: Keeping his family safe, giving them a home, and a place to grow.
Isabella: Satisfying her curiosity. Recognition for her work would be nice as well but she's not going to hold her breath. I really need to get her to adulthood so these answers are more honest. =p
Edward Kenway is currently pretty fixated on money and spiting everyone who ever said that he was too low-born to amount to anything as the hallmarks of success. For him, anyway. He doesn't give a flip about what constitutes a successful life for anyone else. He will be like this for most of the rest of his canon.
I don't think any of them has specific ideas about a successful life as such, but they know what makes or would make their own life successful:
Teja: Getting some of his people to survive him and dying while taking as many enemies with him as he could, after having spent his life buying time for his people and fucking things up for their enemies. Being king was just a necessary and unpleasant step in the process.
Hannibal: Enjoying life to the fullest while not getting caught; that is over now. I think he's in the process of redefining success (rather than admit that he has failed spectacularly).
Father Harman: Keeping humanity safe from vampires, preferably by eliminating the Code Fives, or at least dealing blows to sensitive points in their network.
Lady Margolotta: mainstreaming vampires
Katrina Crane: stopping the apocalypse, then finally making a life of her own with Ichabod while still continuing to fight for the human rights issues of her time -- for her, there will always be a next thing that she has to accomplish before she will declare herself successful.
Tamara the mermaid: Music and good food, preferable both in the shape of easily tractable sailors.
Poins: All debt paid off, and something left to keep. Great success would be a knighthood and an estate, which he knows is never forthcoming, and he would never say so, lest he tarnish his reputation of worldly-wise cynicism.
Lorenzo: Success is what his father wants him to have, that is, political and financial advancement for the family. Lorenzo despises that success and would much rather spend his time with music, painters and people throwing around outrageous ideas. That latter fact, however, historically secured him lasting fame throughout history...
That unnamed potential headvoice I am trying to shut up because my roster is full: Unlimited resources and the ability to do whatever he wants without any consequences whatsoever. Now that he has these, he finds that he is mostly bored out of his skull by the complete lack of conflict or challenge.
Crush your enemies, see them driven before you, and hear the lamentations of the women. Sorry, no, that's not it.
Mark's opinion currently is pretty basic: his family safe, his friends safe, the world as safe as it can be. Money would be nice, but that's already taken care of. Time to goof off, read comics, and see his friends. The ability to not worry about upcoming alien invasions led by his father would be nice, too.
Katara - Supporting and protecting people in need, including and especially her own community.
Asami - Creating something, or contributing to its creation, that lends to the good of the world overall.
Leslie - Using what you have and gain to provide the best you can for others.
Hiccup - Is more at the 'stumbling around and into things' kind of part of life.
Marceline & Manny - Aren't alive and would not consider their 'alive' portions of existence "successful." (Well, Marceline might, because she became a vampire and killed the king, but.)
All Lohengrin cares about to make his life successful is taking as much of the Raven down with him when he dies and ensuring Siegfried and Tutu outlive him.
Mia, having helped secure free will for generations to come, already considers her life quite successful.
Lucas is still figuring it out. If he can make a good world, that'll be enough.
Gene - decent amount of scumbags caught, professional respect from colleagues to acknowledge this.
Javert - well, he did judge it by how irreproachable he was. Now, he has no idea.
Valjean - that it has love in it. And to have used one's time to help as many others as possible.
Bruce Wayne - I think he'll consider himself successful when he's cleaned Gotham up as much as he possibly can, and then managed to find some peace with himself.
Bruce Banner - at the moment, he has no idea. He would have said excellence in his field, but...yeah, the Hulk kind of ruined any chance of that.
Robin - to have earned a living, raised a family, and not have been afraid to fight when needed. Whether it be foreign enemies, or the powerful of his own land.
Courfeyrac - a republican France would have been a great success. As it stands, he still thinks he spent his life wisely. Others will fight on thanks to their deaths, and France will get there eventually. Good enough.
Fairy Fixit wants to be a giant, so that others can stand on her shoulders and see farther, figuratively speaking.
I have no idea what Amascut wants out of life. She has been in a bad, unhappy place for millenia. It's especially bad since she used to have a very good idea and strong opinions about what constituted a worthy and successful life, since she was the Judge of the Dead and would award those who lead worthy lives with rebirth. ;_;
Enjolras: Man, I dunno. If you asked about a satisfying life, or a worthy life, I'd have answers. But a lot of what he's about is rejecting the norms of 'success' (money, marriage, comfortable life within the existing system) of the bourgeois class he came from; I'm not sure he has a tidy definition of success to put in its place. I mean as a broad principle, rather than as a personal definition of "This is what would count as succeeding at what I set out to do." He's satisfied with how his life went, anyway, which is maybe good enough.
Thor: fulfilling the duties which are yours to fulfill; being honorable; being kind where you can.
River: doing what's right. Helping others. Standing up for the greater good. Enjoying life as much as you can, within that.
Regan: doing what's right, providing for yourself and your family, helping others, trying to have as few regrets as possible.
Trowa: Doing the right thing, as much as you can. Protecting others. Not making stupid mistakes -- mistakes happen, and people get unavoidably over their heads sometimes, but the mistakes shouldn't be because you were blind to your own incompetence in some arena. Being happy, if you can; making other people happy, if you can.
Clare: For humans: being happy. For her kind: killing as many yoma as possible, and dying at a friend's hand at the end instead of turning into a monster. (Dying at a yoma's hand only counts if you kill the yoma too in the process, or distract it long enough for a comrade to do so, rather than losing the battle.)
Cosette: loving God and your fellow human beings; giving charity where you can, and helping others as you can; providing for your children, materially and otherwise; doing your best to be a good person.

Madame Thénardier: I lied, she thinks that there is one measure for success for everybody, and that is money. Have lots of money, and the rest of the nice things will automatically come to you. Pretty things, leisure, influence, respect. But money unfairly doesn't come her way, and for that, she secretly blames her husband. Later on, she will mostly, vaguely blame Jean Valjean.
