Every character has a defining moment or quote - may it be directly from canon or spoken by the portrayer or creator of the character in an interview or commentary. What's that one quote you keep in mind when it comes to playing your pups and keeping them IC?
And adding in my own bit, cause some of us come from non-speaking canons: Or what is that one scene or action sequence?

Exibit A: This panel.
Exibit B: "You think you can break me?!? I was born broken."

As to an actual line:
Jim Gordon: Those bastards nearly got you twice, huh? You're quite the survivor.
Selina: You have no idea.

"Why did I waste all my time pretending to be someone I wasn’t? I was tired, so very, very tired of standing on my own brakes. I haven’t felt this good since I lost control and slapped Aunt B. She had backhanded me right down two flights of stairs, but it was worth it. It was so worth it."

"This was now my life. Cinnamon rolls. Sunshine's Eschatology, seeing in the dark, charms that burned into my flesh where I could not lose them. A special relationship with the Special Other Forces, where not everybody was on the same side. A landlady who's a wardskeeper. Untidy closets. Vampires.
Get used to it, Sunshine."

And explanation to what the hell happened to Rae's father and his family.

Will from Paul Creswick's Robin Hood: Stuteley said, in a sad tone: "Sheriff, seeing that I must die today, grant me this one boon, that I may not be hanged upon a gallows tree, but rather that I die with my sword in my hand, fighting you and all your men to the last."
Charles the phrase, "You're not alone." To be that's the linchpin of his character.
Quentin's harder as there are a lot of things but I like this one from Ashes of Honor, "Its what you said we were really good liars." And then lots of great descriptions of him, very hard to pick.
I don't have one of Ivan's canon books handy to pull a quote up, but his wife says that he's warm and that not everyone needs ambition. That captures a lot of why I love him.
The ending of 3:10 to Yuma, link leads to a youtube. Logan Lerman and Christian Bale's acting is fantastic and I think about how much William grows up at the end of the movie.
For Jane, so many scenes but I think this is one of the best moments that captures why I love Jane. I love their faces and there are so many moments I could pick.
For Moist another who I don't have canon with me. “And so we progress. Always keep moving. There may be something behind you.” ~Moist von Lipwig
Sameth, the quote on his journal that he defines himself by making things.
Its hard to pick for Demeter as there's a lot of wonderful stuff out there and I can't pick an exact quote for her.
For Tumnus: "Daughter of Eve from the far land of Spare Oom where eternal summer reigns around the bright city of War Drobe, how would it be if you came and had tea with me?" (LWW, Ch. 2)

Or a bit from the series' protagonist: That's another infuriating thing about him. He's so damned self-confident that he could probably have dinner with Oberon himself and not feel like he was outclassed.

Queen's guard: In the name of the Queen, halt!
Tybalt: In my name, no.

Balthazar: Really, it's when he decides to let Dave off the hook for saving the world, tells him to have a nice life with his girlfriend, and goes to face Morgana alone. But youtube doesn't have a clip of that, so here's a bit with a dragon instead. Dave, you should run.
Oswin: She's been the spotter and last-minute-savior for Team TARDIS ever since they got flung onto the Dalek Asylum... and then the Doctor rips her to bits with a few well-placed words. But she's Oswin Oswald, and she's made of sterner stuff than he imagines - she'll save you too, Chin Boy. And here's a music video for all your owful Oswin needs.
Clara: Two stand out for me (just take all of the poor life choices as given, honestly, this girl takes danger as an invitation ON ACCIDENT), but still, two: One, when she offers a parasite god something she holds deeply precious to save a man she barely knows and a planet that won't exist until far in her future, and one that hasn't ended up on Youtube yet, where Clara gets fed up with Madam Vastra's insinuations and lays out a verbal smackdown. She might be a tiny human, but she's learned psychological warfare from living with a master of the art. It's in 'Deep Breath', which is an awesome episode.
Glorfindel: It wasn't part of the movies (something I'm still sad about), but it was a scene I've re-read so many times I can almost quote it from memory. It's when the hobbits and Aragorn have finally found the road again during their flight from Bree to Rivendell, and Frodo's fading fast. The world is dark and full of danger, and the hobbits are exhausted and running out of hope.
...
And then this shining white figure on a white stallion with bright silver bells that ring merrily sewn into the tack comes galloping down the road, utterly unafraid and fully of joy. That's him, not just defiant of the world's darkness but exultantly so.
Sam Stewart: Also fails to make it on Youtube (most of the clips there are either in music videos shipping Foyle/Sam, or of the newest season that I'm trying not to spoil myself more as I'm still in season 5), but one of the first scenes to point out how Sam's going to be is in the first episode - Sam's left alone with the car while Foyle goes to hunt a man responsible for helping draft dodgers. ... And Sam's the one who catches the fellow, smashing him over the head with a rubbish lid. She's steady and brave and unconventional and filled with good attitude when the world's going to hell in a handbasket. Here, have a clip of Sam trying to use a typewriter. It's just as well she never wanted to be a secretary.
Katya: I think the best scene for her is early on - Anton has found this boy that he's pretty sure has something to do with the pack of vampires he failed to put down earlier in the book (and it turns out those vampires are part of a much larger plot to disrupt the balance between the Watches), but now he's being called to help deal with a gal that might be accidentally be causing the end of the world. So the boss sends his two heavy hitters (Bear and Tiger Cub) to guard the boy. Katya (Tiger Cub) shows up full of energy and cheerfulness and complete (at least superficially) disregard for the seriousness of the situation, and by the time she's done the boy's family has taken her and Bear in as long lost cousins from some outer district who have come to visit and they're having a party. Seriously. Why just sit around and guard when you can have a party.
Bones: This. That whole episode, actually. Oh Bones.
Jemma: This whole episode. The one I spent flailing at the TV, yep. Warning, spoilers for FZZT and all that implies, because holy heck they didn't hold back in that episode.
EDIT: And this for Jemma too, because yes, she's brave and clever and all of that but sometimes the voice that goes inside the head and the voice that goes outside the head get messed up and really she didn't mean to say all of that out loud. In public. Oh geeze...

Alanna: So? He was stupid. If I killed everyone who was stupid, I wouldn't have time to sleep.

"Doesn't matter what the press says. Doesn't matter what the politicians or the mobs say. Doesn't matter if the whole country decides that something wrong is something right.
"This nation was founded on one principle above all else: the requirement that we stand up for what we believe, no matter the odds or the consequences.
"When the mob and the press and whole world tell you to move, your job is to plant yourself like a tree beside the river of truth, and tell the whole world--"
"'No. You move.'"

He says this in between gunfights during his and his friends' Grand Last Stand. To be fair: there is an actual cat inspiring the comment. To be accurate: He'd probably say it anyway. Joly, you nerd.

That's her, for me. The awareness, the innocence and the kind of oh-gosh-I-didn't-know-that'd-work-so-well humility and grace she has. And the thread of serenity and the wonder, through it all. ... Ah-hem.

Clementine: "Clementine doesn't care. She'll just shove it in your face." - Wendi McLendon-Covey, Season 2 commentary track.
Dixie: "Marriage is a wonderful institution, but I don't want to be committed just yet."
Pinkie: "Oh, Oatmeal, you're hilarious!"
Juliet: The line I can't quote offhandedly about getting Christmas cards from people she's arrested.
Eponine: "He was never mine to keep."

I LOVE Dixie's quote. =]
