Connor hasn't gotten there quite yet in Barplay, but I imagine he'd be pretty stoked to earn the proper-noun-Assassin robes. In-game it's just a bunch of tutorial missions and you're done, but I imagine it'd take a few years of some pretty strenuous mental and physical conditioning. Think Yoda and Luke in the swamp, only he won't be feeling out the force so much as examining classical philosophy in-between conditioning sessions ripped from a Rocky montage.
B.J would be very fond of the time his dad praised him for how he quelled a cattle stampede during a thunderstorm.
Maverick would probably go with earning his aviator's wings. Because he's got daddy issues. In the sense that he's constantly measuring himself against his dead father - or rather, his mental construction of his dead father that probably elevates dear old dad into something he wasn't. Yes.

For a scientist, there's not always a single moment. A 'Eureka!' moment. There's a lot of research, testing, failure, testing again. More research. For Dejah, the closest I can get is the day she figured out that the Thern really had sabotaged her experiments. That she was on the right track from the very beginning and they actively thwarted her work. (And when that didn't work, they tried to have her assassinated.)
That was the moment when she knew they were going to win.
Cassie's would be her Q Course graduation. (Or whatever her universe's equivalent is - I'm operating on the assumption that the Army in the MK-verse somewhat resembles the RL one behind the scenes. As such, I'm probably giving more thought to the details than the writers did).
Garyn isn't really proud of much. You'd probably have to force something out of him, at which point he'd mutter something about saving a comrade's life on a job in Leyawiin or something.

Sheogorath isn't sure, but it probably isn't the time he filled a pit up with clowns to try and stop the Greymarch.

Tavi: Right now, in-Bar, holding the Elinarch against impossible odds might still be it, although rescuing the remnants of the Canim might be in the running. Honestly, though, very shortly it'll change to two easily definable moments. One he can't take credit for, being when his first child is born. The other will be the first morning he is officially, fully confirmed First Lord and his first major action ends slavery in Alera. Just signing a paper, but it's a drastic change (one his family--both sides--have fought for since before he was born) and kind of a symbol of the first of many, many changes. Ultimately he can't have a single moment because when he's done, of course, but that one summarizes it.
(No, stopping the apocalypse doesn't actually make the list. He stopped it, and it's the thing buying him more power than he'd normally carry politically, but it isn't a point of pride. Somehow.)
And now I think I want Tess and Hank to meet, being Harvard allum and all. Of course I haven't decided exactly what he graduated from Harvard with, canon only states "he graduated from Harvard at 15".
