Swamp Thing's mortal enemy - or perhaps immortal enemy - is Anton Arcane. Whenever there is a Swamp Thing comic, Arcane is just around the corner.
Cyborg has something of a millicanon rivalry with Jinx, but on the cartoon his enemies are the Titans' enemies. And there are a lot of them. I suppose Brother Blood would be at the top of Cy's list, but he wasn't that interesting a villain.
Poirot was once challenged to solve "The ABC Murders," and played dead once, but even though he has sent many to prison or the gallows, most are not really out to get him in particular. That wasn't how things worked in Christie's world.

Carmen Sandiego isn't so much out to get the Chief as she is out to get the Statue of Liberty, Lake Baikal, the Rio Olympics (all of it), the royal palaces of Abomey, Stonehenge, the world population of Tasmanian devils, and McMurdo Station.

There have been a number of people and organizations who have, in the past, been out to get Ellen. Pretty much all of them are dead now.

Shephard has dealt with most of his enemies and does not worry about the others too much. Keep an eye out for alien monsters, surviving Combine, and the Butler brothers, who will eventually be tracked down and given a very public trial before a very public and proper execution because some genes don't belong in the pool, that's how he sees it.
Santo has a couple of different enemies; I think he's probably most concerned about the individual called El Rey del Crimen, but I haven't seen that part of his canon yet so I don't know what level of threat the King of Crime poses.
Quicksilver does not have anyone out to get him and anyway if he did he would laugh at them as he left them in his dust. And then probably pants them when they weren't looking.
Stacker Pentecost has his suspicions about the sterling specimens of humanity on the World Council but does not really think they are out to get him.
Whenever YT dons her Kourier uniform and goes to work, she adopts the mindset that just about everyone is out to get her - truckers, cops, luxury car drivers, 'roided-up young Studleys behind the wheel of their family minivan. So she does everything she can to keep would-be adversaries off balance and drive home the message that she is not an easy target. Just look at the canon quote in her journal profile!
Arguably, YT's attitude is mostly justified, given that many people in her world view Kouriers as minions who deliver the mail at best and troublesome subhuman scum at worst.

Kyouko is very much Hermonie Granger as a popular/mean girl. Suzuka might be out to get Touji but more as collateral damage to Harutora.
Selina: Gotham PD and Alfred. Both are very much out to get her, though Alfred is more interested in protecting Bruce than harming Selina.
Amelia: Her mother and yes, it is very real and not in Amelia's head at all.
Eliot: Total head canon, but I am imagining the current head of the physical kids house is out to get Eliot.
Hank: Magneto perhaps, though that's only if he gets in Magneto's way. The general population, if you believe in mob fear of mutants.
Which leaves Rollo. Currently, I'd have to say King Horik but truthfully, it's Rollo. Ragnar sort of is, but that is really in Rollo's head.

The Crows are after her and her beloved Zevran, the Wardens at Weisshaupt are not on her side, Howe's supporters still hate her, so do swathes of still-nonsentient Darkspawn and probably some of the sentient ones, the House of Repose in Orlais might have some nobles that want to take her out as a way to take certain talking points off the table and to remove one of Alistair's supporters, Chantry-folk of certain persuasions hate the Fereldan reworking of the Circles and that is largely her fault . . . .

Dean Winchester has demons, angels, and almost every supernatural creature out there, plus assorted hunters and the FBI on his tail. Most of those are trying to kill him. Good times!
Sunshine has dealt with any master vampires out to get her at the moment (as far as she knows), but she knows very well that Jain, the Goddess of Pain, the Deputy Executive Director of Special Other Forces, is out to get her. As a 'well-meaning and civic-minded' person in a position of power, the Goddess of Pain has reason to suspect Rae Seddon, neighborhood baker, has been fraternizing with enemy Others, has lied to and withheld evidence from SOF interrogators, and is a danger to the public's safety.
Emcee: lololol, you know, those guys. That's at home, but in Milliways he's become a lot more paranoid, partly because of his own situation of having to constantly hide and be on guard, and the fact that he doesn't consider the main bar to be a safe space anymore. (Thanks, flying cameras!) Anything he says can be used against him or someone he cares about, and it's not really who he knows might get him, but who he doesn't know.
