My first played was Sybil Trelawny from Harry Potter. My first apped (and wasn't approved until the pilot actually aired, but I had a copy of the uncut pilot before that, *cough*) and stuck around the longest was Charlie Pace from Lost. I played him for ... gosh, five years, maybe? A long time, anyway. Sybil was kind of one-note and I didn't see many places to go with her at the time.
I had intended to app the Fool from Robin Hobb's Assassin's/Liveship/Fool's trilogies, and I sought out Milliways specifically for that purpose after seeing it some months previous when Oz-from-Buffy's player had asked for writing critique.
Anyway, the Fool was taken, and I'd played KOTOR 2 recently, so it ended up being Atton, who is Not like the Fool, instead.
Aradia Megido, who's still on my roster because I haven't felt the need to drop anyone yet and the voice is still present albeit rusty from disuse, although I don't really feel confident bringing her back properly without being caught up on what the other Homestuck players did after I dropped off the face of the earth in 2014.
For me, it was Emperor Joshua Norton the First. I chose the Sandman short Three Septembers and a January for his canon. He was a lot of fun but ultimately was limiting for me. I still get tempted from time to time to play him in a meta kind of way; giving him access to any canon using his personage via Milliways' front door.
